I've probably seen this meme/joke a million times but to this day I still don't get it. Is it the sheer ridiculousness of asking a BB&B for a Wizard killing silver bullet or am I missing something?

 

I think the joke is that at first you think it's just a misunderstanding where an employee (reasonably) assumes he wants a blender, but he actually wants a wizard killing bullet (plus the oddity of seriously asking for a magical anything). But then your expectations are subverted because the employee can help him find his magical ammunition.

 

The joke is that it appears to be a completely vanilla situation, but there's a hidden fantasy/spacelizard reality hidden underneath. It's like, say the password to the owner of Spellbookse Moste Raree and get access to the super secret stash of mega items. Isn't that straight from an RPG?
